چکیده

Immanuel Kant noted this approach would not be suitable for understanding complex systems such as an organism. He said there would never be a " Newton of the blade of grass " : because the blade of grass is more than the sum of its parts, complete knowledge of the parts list will not necessarily be predictive of the behavior of the whole. Fortunately, we don't have to resort to a vitalistic explanation for the phenomenon of emergence. Systems biology offers both an epistemological and technological solution to this problem. Ideker et al. have outlined the four steps of the systems biology approach: component identification and modeling; system perturbation and monitoring; model refining; and model testing. In addition to high-throughput monitoring and reverse-genetics tools, the volume and nature of the data now being generated necessitate the integration of computational modeling and testing. The basic premise is that the identification of a parts list, combined with the monitoring and modeling of the dynamic behavior of those parts, will offer better models of biological systems. The challenge of systems biology. The application of a systems theoretic analysis may dramatically improve our understanding of many biological processes such as morphogenesis, pathogenesis, cognition and ecology. There is no question that these all may be complex systems that preclude full understanding by the reductionistic paradigm. But this new incarnation of systems biology faces a number of challenges in the coming years. The first is technological: to move beyond a focus on transcriptional profiling and protein–protein interactions. A more complete understanding of system dynamics requires knowledge from other parts of the system, such as metabolites, as well as other forms of network modulation, such as post-transcriptional regulation. The second, and perhaps most vexing hurdle, is data sharing. If different datasets are to be integrated in a meaningful way, it will be necessary to not only improve annotations, but also to increase standardization in lab procedures. It is axiomatic in molecular biology that minor variations in procedures, reagents or environment can have a dramatic effect on biological systems. It is thus incumbent on systems biology researchers to communicate more effectively about system parameters that may be relevant to the ultimate model. Lastly, this approach requires expertise in molecular biology, mathematics, genomics, computer science and systems theory. As it is improbable that effective progress in the field will be made by individual researchers with all of these skills, …

Evolutionary and Ecological Genetics of Medicago truncatula

This chapter synthesizes our current knowledge on the ecological genetics of the model legume Medicago truncatula. After a short introduction to the life cycle of the species, we describe the mating system of M truncatula and how selfing rates in nature can be inferred both by progeny testing and population surveys. We then review what is currently known on the distribution of genetic diversity...

Comparative Analysis of the Symbiotic Efficiency of Medicago truncatula and Medicago sativa under Phosphorus Deficiency

Phosphorus (P)-deficiency is a major abiotic stress that limits legume growth in many types of soils. The relationship between Medicago and Sinorhizobium, is known to be affected by different environmental conditions. Recent reports have shown that, in combination with S. meliloti 2011, Medicago truncatula had a lower symbiotic efficiency than Medicago sativa. However, little is known about how...

Genome-Wide Identification and Expression Analysis of the 14-3-3 Family Genes in Medicago truncatula

The 14-3-3 gene family, which is conserved in eukaryotes, is involved in protein-protein interactions and mediates signal transduction. However, detailed investigations of the 14-3-3 gene family in Medicago truncatula are largely unknown. In this study, the identification and study of M. truncatula 14-3-3-family genes were performed based on the latest M. truncatula genome. In the M. truncatula...

The model legume Medicago truncatula A17 is poorly matched for N2 fixation with the sequenced microsymbiont Sinorhizobium meliloti 1021.

Medicago truncatula (barrel medic) A17 is currently being sequenced as a model legume, complementing the sequenced root nodule bacterial strain Sinorhizobium meliloti 1021 (Sm1021). In this study, the effectiveness of the Sm1021-M. truncatula symbiosis at fixing N(2) was evaluated. N(2) fixation effectiveness was examined with eight Medicago species and three accessions of M. truncatula with Sm...

Chromosomal and symbiotic relationships of rhizobia nodulating Medicago truncatula and M. laciniata.

Multilocus sequence typing (MLST) is a sequence-based method used to characterize bacterial genomes. This method was used to examine the genetic structure of Medicago-nodulating rhizobia at the Amra site, which is located in an arid region of Tunisia. Here the annual medics Medicago laciniata and M. truncatula are part of the natural flora. The goal of this study was to identify whether distinc...

Comparison of rhizobia that nodulate Medicago laciniata and Medicago truncatula present in a single Tunisian arid soil.

The rhizobia present in a single arid region Tunisian soil that nodulate Medicago laciniata and Medicago truncatula were compared. All isolates, 40 from each host, were Sinorhizobium meliloti based on 16S rRNA polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) patterns and subsequent confirmation by sequence analysis of the 16S rRNA genes in four representatives from ...
